well, first of all, do you see more than one hard drive icon on the desktop? if not then there are other ways. the best way to see if there is more than one is to open the mac and count them. but there is a way within the finder. goto the preferences under the finder menu. in the resulting diag box, make sure that hard drives is checked. now all the formated hard drives that the mac sees will show on the desktop. if only one shows up, then the other may be bad, or unformatted. goto your utilities folder in the applications folder and find an app named "disk utility" and launch it. on the left side it will list all the drives the the mac sees. it will list the drives with model number, and the volumes for each drive right below the drive. see if there is a drive without any volumes, or with volumes that you do not see on the desktop. if the only one that is shown is the one you do know of, then you will have to open the mac and see if there is anymore drives, because if it doesn't show up in the utility, then its bad, or not plugged in. of course there is always the option that your boss doesn't know what he's talking about, because the last employee may have been using an external drive that they only plugged into one of the firewire, or usb ports to do the backup, and took the drive with them when they left.
