Among others, I have this beautiful icon..

..That I want to apply to a certain folder, but don't know how. If I copy and paste it, it isn't transparent or semitransparent in spots, but instead, black.

How Do I do it?

Use img2icns. It's free, and it will take an image with transparency, like a png or psd, and create a nice icon out of it.

The main downside to img2icns is that it does not let you specify different images for different sizes. For that, you could install Apple's developer tools (available on your Mac OS X installation DVD, or as a free download) and use their own app called Icon Composer. If you don't demand that level of control, though, img2icns is just fine.
 
Oh, you mean it's already a .icns file? In that case, try IconDroplet. It will take icns files and make Finder icons out of them. After that you can copy the icon in the Finder and then paste it onto other items.
