mail will not open except with a warning

my mail opens with warning "home folder is full - make space, etc." with the only option being "quit"
I have not been able to open the program for three weeks.
I have tried;
contacting the local isp
shutting computer on and off
using "help" feature on my Mac, etc.

Have you tried following the suggestion of the error message, and freeing up some space on your hard drive from your home folder?

Also, what is the exact wording of the error message (I doubt the error message says specifically, "make space, etc.")? Error messages can be easily misconstrued and changing a single word can change the entire meaning of the message. Please quote the message verbatim, as it appears on your screen.
 
First all restart your Mac. Next download either Yasu or Onyx and run all the "cleaning routines". Let the program restart your Mac and then manually restart again to completely rebuild your Shutdown/Startup cache.

See if that helps. Plus how much space is left on your Mac's hard drive? What version of OS X are you running on what Mac?
